Acid‐Cleavable Poly(ethylene glycol) Hydrogels Displaying Protein Release at pH 5

Abstract PEG is the gold standard polymer for pharmaceutical applications, however it lacks degradability. Degradation under physiologically relevant pH as present in endolysosomes, cancerous and inflammatory tissues is crucial for many areas. The authors present anionic ring‐opening copolymerization of ethylene oxide with 3,4‐epoxy‐1‐butene (EPB) and subsequent modification to introduce acid‐degradable vinyl ether groups as well as methacrylate (MA) units, enabling radical cross‐linking. Ring splitting of azetidin-2-ones via radical anions

The radical anions of azetidin-2-ones, generated by UV-irradiation in the presence of triethylamine, undergo ring-splitting via N-C4 or C3-C4 bond breaking, leading to open-chain amides. This reactivity diverges from that found for the neutral excited states, which is characterised by alpha-cleavage. The preference for beta-cleavage is supported by DFT theoretical calculations on the energy barriers associated with the involved transition states. Thus, injection of one electron into the azetidin-2-one moiety constitutes a complementary activation strategy which may be exploited to produce new chemistry.

Hemoglobin loaded polymeric nanoparticles: preparation and characterizations.

In the present work polymeric nanoparticles based on Poly (maleic anhydride-alt-butyl vinyl ether) 5% grafted with m-PEG (2000) and 95% grafted with 2-methoxyethanol (VAM41-PEG) were loaded with human hemoglobin (Hb) and characterized from a physicochemical point of view. The assessment of structural and functional features of the loaded Hb was performed and the effect of the introduction of different reducing agents as aimed at minimizing Hb oxidation during the nanoparticles formulation process, was also investigated. Semi-IPN's of different composition, based on linear poly(methyl vinyl ether) (PVME) and crosslinked polystyrene are studied using stress and birefringence measurements in simple elongation. From quasistatic stress-strain experiments at constant T(experiment)-Tg, it can be deduced that the stress results only from the deformation of the polystyrene network, while the PVME chains do not contribute to the macroscopic stress. Density functional theory study of the Lewis acid-catalyzed Diels-Alder reaction of nitroalkenes with vinyl ethers using aluminum derivatives

The inverse electron demand Diels–Alder reaction of nitroalkenes with vinyl ethers catalyzed by Lewis acids based on aluminum metal, AlMe3, Al(OMe)3 and AlCl3 was studied using density functional theory methods. A continuum model was selected to represent the effects of dichloromethane used as solvent. For this cycloaddition reaction two reactive channels corresponding to the endo and exo approach modes of the vinyl ether to the Lewis acid-coordinated nitroalkene were studied. Coordination of the aluminum metal to an oxygen atom of the nitroalkene increases the electrophilicity of the heterodiene, decreasing the activation energy of the cycloaddition. Cleavable Polyethylene Glycol: 3,4-Epoxy-1-butene as a Comonomer to Establish Degradability at Physiologically Relevant pH.

Polyethylene glycol (PEG) has been used for decades to improve the pharmacokinetic properties of protein drugs, and several PEG-protein conjugates are approved by the FDA. However, the nondegradability of PEG restricts its use to a limiting molecular weight to permit renal excretion. In this work, we introduce a simple strategy to overcome the nondegradability of PEG by incorporating multiple pH-sensitive vinyl ether moieties into the polyether backbone. Continuous polymer fractionation of poly(methyl vinyl ether) and a new Kuhn-Mark-Houwink relation

Continuous polymer fractionation (CPF) — a new large-scale method — was applied to the technical poly(methyl vinyl ether) (PVME) using toluence as solvent and petroleum ether as non-solvent. Two different devices served as fractionation apparatus, namely a column packed with glass beads and a set of mixer-settler extractors. Advantages and draw-backs of these devices are discussed. The experiments yielded numerous PVME fractions, varying in molar mass from 22 to 87 kg/mol and in molecular non-uniformity U = (M¯w/Mn) – 1 from 0,25 to 0,41 according to gel-permeation chromatography as compared with ca. Poly ((ethylene oxide)-b-(propylene oxide)-b-(ethylene oxide)) triblock copolymers commonly known as poloxamers or Pluronics constitute an important class of nonionic, biocompatible surfactants. Here, a method is reported to incorporate two acid-labile acetal moieties in the backbone of poloxamers to generate acid-cleavable nonionic surfactants. Proteins represent a versatile biopolymer material for the preparation of nanoparticles. For drug delivery applications an acid-triggered disassembly and payload release is preferred. Herein, we present a protein nanoparticle system based on cytochrome c, which is surface-modified with acid-degradable polyethylene glycol (PEGylation). pH-Sensitivity was obtained through vinyl ether moieties distributed in the polyether backbone.
